Complexometric Titration

การไทเทรตสำหรับปฏิกิริยาการเกิดสารเชิงซ้อน (Complexometric titration)*
หลักการของ complexometric titration เป็นการไตเตรทเพื่อหาปริมาณ metal ion โดยการเกิดเป็นสารประกอบเชิงซ้อนระหว่างไอออนของโลหะ ซึ่งเป็นตัวรับอิเลคตรอนกับสารที่สามารถให้คู่อิเลคตรอนกับโลหะได้ ในสารละลายที่ถูก buffered ให้มี pH อยู่ในช่วงที่เหมาะสม อินดิเคเตอร์ที่ใช้จะถูกเติมลงไปในสารละลายนี้ แล้วไอออนของโลหะจะถูกไตเตรทด้วยสารละลายมาตรฐานของ complexing agent และเมื่อถึงจุดยุติควรสังเกตเห็นการเปลี่ยนแปลงสีได้ชัดเจน การตรวจวัดจุดยุติอาจทำได้ทั้ง physical method และ chemical method การไตเตรทโดยวิธีนี้จะทำได้ง่าย สะดวกและมีความถูกต้อง ดังนั้นจึงได้มีการพัฒนามาใช้แทนการวิเคราะห์โดย gravimetric method ซึ่งวิเคราะห์ไอออนโลหะได้หลายชนิด

    * หลักการนี้ ไม่สามารถใช้วิเคราะห์โลหะ alkali metal ได้
    * Analyte: metal ion ที่มี valency > 1 เช่น Al3+, Cu2+
    * Titrant: Chelating agent หรือ complexing agent หรือ Ligand เช่น EDTA
    * Complex : Chelate
    * การวัดจุดยุติ : Indicator หรือ potentiometric method

A technique of volumetric analysis in which the formation of a colored complex is used to indicate the end point of a titration. Also known as chelatometry. Also spelled compleximetric titration.

Complexometric titration (sometimes chelatometry) is a form of volumetric analysis in which the formation of a colored complex is used to indicate the end point of a titration. Complexometric titrations are particularly useful for the determination of a mixture of different metal ions in solution. An indicator capable of producing an unambiguous color change is usually used to detect the end-point of the titration.

การจะเลือกใช้ complexometric titration method ในการวิเคราะห์หาปริมาณโลหะชนิดต่าง ๆ ได้นั้นต้องพิจารณาถึงหลักเกณฑ์ดังนี้
1. Complex reaction ต้องเป็น stoichiometrically กล่าวคือ metal ion ต้องทำปฏิกิริยากับ chelating agent ในอัตราส่วนที่แน่นอน
2. อัตราการเกิดปฏิกิริยาต้องเร็ว
3. สารประกอบเชิงซ้อนที่เกิดขึ้นต้องมีความคงตัวดี ค่าคงที่ของการเกิดสารประกอบเชิงซ้อนต้องสูง
4. มี sharp end point กับอินดิเคเตอร์ คือสังเกตจุดยุติได้ชัดเจน เมื่อเติม titrant excess ปริมาณเล็กน้อย titrant นั้น ก็สามารถทำปฏิกิริยากับอินดิเคเตอร์แล้วเกิดการเปลี่ยนแปลงสีที่ชัดเจน
5. ไม่เกิดตะกอนระหว่างการไทเทรต เพราะจะทำให้สังเกตเห็นปฏิกิริยา หรือสีของจุดยุติได้ไม่ชัดเจน

Ligand, Chelating Agent, Complexing Agent
- โมเลกุลที่เป็นกลางหรือมีประจุลบที่สามารถให้คู่ electron กับ central atom
- โมเลกุลหรือสารที่ประกอบด้วยอะตอมอย่างน้อย 1 อะตอมที่มีอิเล็คตรอนคู่หนึ่งเป็นอิสระ (unshared pair of electron) สามารถให้อิเล็คตรอนกับโลหะได้ ligand อาจเป็นโมเลกุลที่ไม่มีประจุเช่น แอมโมเนีย หรือเป็นประจุลบ เช่น เฮไลด์ ไฮดรอกไซด์ ซัลเฟตและฟอตเฟต เป็นต้น
- ในโมเลกุลมีอะตอมของธาตุที่สามารถให้คู่ electron ได้ เช่น O, N, X (halogen) และ S เป็นต้น
- -NH2 และ –COO- เป็นหมู่ที่ให้ e- ได้ดี
- Lewis base

แบ่ง ligand ได้ตามจำนวนหมู่ที่ให้อิเล็คตรอนอิสระคู่หนึ่งซึ่งมีอยู่ใน 1 โมเลกุลของ ligand ที่สามารถจับกับโลหะ
1. Unidentate or Monodentate Ligand เป็น ligand ที่มีหมู่ซึ่งให้อิเล็คตรอนอิสระเพียงคู่เดียวใน 1 โมเลกุล ตัวอย่างเช่น เฮไลด์ อิออน (Cl-, I-), CN- และแอมโมเนีย (NH3), H2O
ลักษณะการจับกันระหว่าง unidentate ligand กับโลหะที่มี coordination number 6 มีโครงสร้างเป็น octahedral        

2. Bidentate ligand เป็น ligand ที่มีหมู่ซึ่งให้อิเล็คตรอนคู่หนึ่งซึ่งไปจับกับโลหะอยู่ 2 หมู่ใน 1 โมเลกุล ตัวอย่างเช่น ethylenediamine, ethylenediphosphine(diphos), bipyridyl(bipy)

Bidentate ligands have two lone pairs, both of which can bond to the central metal ion. The two commonly used examples are 1,2-diaminoethane (old name: ethylenediamine - often given the abbreviation "en"), and the ethanedioate ion (old name: oxalate).

3. Multidentate ligand หรือ polydentate ligand เป็น ligand ที่ใน 1 โมเลกุลมีหมู่ที่ให้ อิเล็คตรอนอิสระคู่หนึ่งมากกว่า 2 หมู่ ถ้า ligand มีหมู่ซึ่งให้อิเล็คตรอนอิสระคู่หนึ่งอยู่ 3 หมู่ใน 1 โมเลกุลเรียกเป็น tridentate ligand ส่วน ligand มีหมู่ซึ่งให้อิเล็คตรอนอิสระคู่หนึ่งอยู่ 4 หมู่ใน 1 โมเลกุลเรียกเป็น quardridentate ligand

A quadridentate ligand
A quadridentate ligand has four lone pairs, all of which can bond to the central metal ion. An example of this occurs in haemoglobin (American: hemoglobin). The functional part of this is an iron(II) ion surrounded by a complicated molecule called heme. This is a sort of hollow ring of carbon and hydrogen atoms, at the centre of which are 4 nitrogen atoms with lone pairs on them.
Heme is one of a group of similar compounds called porphyrins. They all have the same sort of ring system, but with different groups attached to the outside of the ring.
Each of the lone pairs on the nitrogen can form a co-ordinate bond with the iron(II) ion - holding it at the centre of the complicated ring of atoms. The iron forms 4 co-ordinate bonds with the haem, but still has space to form two more - one above and one below the plane of the ring. The protein globin attaches to one of these positions using a lone pair on one of the nitrogens in one of its amino acids. The interesting bit is the other position.  Overall, the complex ion has a co-ordination number of 6 because the central metal ion is forming 6 co-ordinate bonds. The water molecule which is bonded to the bottom position in the diagram is easily replaced by an oxygen molecule (again via a lone pair on one of the oxygens in O2) - and this is how oxygen gets carried around the blood by the haemoglobin. When the oxygen gets to where it is needed, it breaks away from the haemoglobin which returns to the lungs to get some more. You probably know that carbon monoxide is poisonous because it reacts with haemoglobin. It bonds to the same site that would otherwise be used by the oxygen - but it forms a very stable complex. The carbon monoxide doesn't break away again, and that makes that haemoglobin molecule useless for any further oxygen transfer.

A hexadentate ligand
A hexadentate ligand has 6 lone pairs of electrons - all of which can form co-ordinate bonds with the same metal ion. The best example is EDTA. EDTA is used as a negative ion - EDTA4-

Fajans method เป็นหนึ่งในการไทเทรตโดยวิธีทำให้ตกตะกอนในรูปของเกลือของซิลเวอร์ หรือที่เรียกว่า Argentimetry

In the Fajans method, typically dichlorofluorescein is used as an indicator; the end-point is marked by the green suspension turning pink. Prior to the end-point of the titration, chloride ions remain in excess. They adsorb on the AgCl surface, imparting a negative charge to the particles. Past the end-point, excess silver(I) ions adsorb on the AgCl surface, imparting a positive charge. Anionic dyes such as dichlorofluorescein are attracted to the particles, and undergo a color change upon adsorption, representing the end-point. Eosin (tetrabromofluorescein) is suitable for titrating against bromide, iodide, and thiocyanate anions, giving a sharper end-point than dichlorofluorescein. It is not suitable for titrating against chloride anions because it binds to AgCl more strongly than chloride does.

Adsorption indicators can be used in many precipitation titrations, not just argentometric methods. Let’s imagine that we wish to analyte Cl− in a sample solution by titrating with Ag+; the titration reaction would be Ag+ + Cl− <-> AgCl(s)
Silver chloride forms colloidal particles. Before the equivalence point, the surface of the precipitant particles will be negatively charged due to the adsorption of excess Cl− to the surface of the particles. A diffuse positive counter-ion layer will surround the particles. When the equivalence point is reached, there is no longer an excess of analyte Cl−, and the surface of the colloidal particles are largely neutral. After the equivalence point, there will be an excess of titrant Ag+, some of these will adsorb to the solid AgCl particles, which will now be surrounded by a diffuse negative counterion layer. Adsorption indicators are dyes, such as dichlorofluorescein, that usually exist as anions in the titration solution. The doubly charged dichlorofluoroscein anion is attracted into the counterion layer immediately
following the equivalence point, when the surface charge of the particles changes from negative to positive. For reasons that are not fully understood, the closer proximity of the dye to the particles changes the color of the molecule, providing a visual indication of the titration endpoint. In the case of dichlorofluorescein, the indicator changes to a pinkish color.

Fajan’s Method 
หลักการคือ ที่จุดยุติ ตะกอนจะดูดซับอินดิเคเตอร์ไว้เกิดการเปลี่ยนแปลงสีของตะกอน โดยใช้Fluorescein เป็นอินดิเคเตอร์

ปัจจัยที่มีผลต่อการไทเทรต
- Indicator ต้องแตกตัวที่ pH ที่ทำการทดลอง
- พื้นที่ผิวของตะกอน ควรเติม dextrin, gelatin agar, PEG 400
- แสงสว่าง ทำให้สารสลายตัวง่าย
- การคนหรือการเขย่า ไม่ควรเขย่าแรงเกินไป

BEAKER
บีกเกอร์ เป็นแก้วใส ใช้สำหรับบรรจุสารที่มีปริมาณมาก เพื่อละลายสารหรือทำปฏิกิริยาเคมี และสามารถเทสารออกได้ง่ายทางปากบีกเกอร์ โดยจะมีขีดบอกปริมาตรซึ่งเป็นค่าโดยประมาณเท่านั้น

บีกเกอร์มีหลายขนาดและมีความจุต่างกัน โดยที่ข้างบีกเกอร์จะมีตัวเลขระบุความจุของบีกเกอร์ ทำให้ผู้ใช้สามารถทราบปริมาตรของของเหลวที่บรรจุอยู่ได้อย่างคร่าวๆ และบีกเกอร์มีความจุตั้งแต่ 5 มิลลิเมตรจนถึงหลายๆลิตร อีกทั้งเป็นแบบสูง แบบเตี้ย และแบบรูปทรงกรวย (conical beaker) บีกเกอร์จะมีปากงอเหมือนปากนกซึ่งเรียกว่า spout ทำให้การเทของเหลวออกได้โดยสะดวก spout ทำให้สะดวกในการวางไม้แก้วซึ่งยื่นออกมาจากฝาที่ปิดบีกเกอร์ และ spout ยังเป็นทางออกของไอน้ำหรือแก๊สเมื่อทำการระเหยของเหลวในบีกเกอร์ที่ปิดด้วยกระจกนาฬิกา (watch grass) 

A beaker is a simple container for stirring, mixing and heating liquids commonly used in many laboratories. Beakers are generally cylindrical in shape, with a flat bottom and a lip for pouring. Many also have a small spout to aid pouring. Beakers are available in a wide range of sizes, from one millilitre up to several litres.

FLEAKER
A fleaker is a type of container for liquids used in the laboratory. It can be described as a cross between the Griffin beaker and the Erlenmeyer flask.

Like a beaker, the bottom is flat, with the sides meeting the bottom at a 90 degree angle. The sides are vertical for most of the height; near the top, the sides curve in to form a neck with a widely flared rim. The wide rim makes it easier to pour from or filter into; the narrow neck reduces loss of the contents due to splashing and serves as a grip for handling and pouring. Fleakers have a plastic lid with a built in rubber stopper. When on the fleaker, the lid covers the narrow neck. Fleakers work as well as other glassware for liquids and solutions, but are inappropriate for slurries, precipitates, and recrystallizations (since the narrow neck makes it difficult to remove solids completely from a fleaker).

The fleaker was invented by Roy Eddleman, founder of Spectrum Medical Industries (now Spectrum Laboratories).
